Illinois vs. Duke Picks and Predictions, Saturday, September 6, 2025

By: Nathan Smith Published 09/05/2025, 12:13 AM ET

The College Football season is off and running, and after some fantastic, high-scoring, back-and-forth battles a week ago, we'll hope for another this week as we take a look at our Illinois vs. Duke prediction.

The 11 Illinois Fighting Illini (1-0) made a statement in week one, and with the polls already having them ranked No. 11, this is a team that's ready to make another statement on the national stage in this matchup.

On Saturday, they take on the Duke Blue Devils (1-0), a team that's been good in recent times, but after struggling against elite-level talent, this will be a massive test for them early in the 2025 campaign. Read on to see our Illinois vs. Duke prediction.

When it comes to college football selections, our NCAAF Predictions are stellar.

Can Illinois make another statement this week?

The Illinois Fighting Illini came into the 2025 season ranked No. 12 in the AP Polls, and after a dominant home win in week one, they've done nothing to shake that confidence, as they have now climbed one spot to be No. 11 heading into week two. This comes off the back of a 10-3 record in 2024 and a major bowl win over South Carolina, and with Luke Altmyer back for his senior campaign, the stars could be aligning for a massive season.

In week one, the team dominated on both sides of the ball, with Altmyer throwing for 217 yards and three scores as the team managed a very balanced 440 yards on the day. In the victory over Western Illinois, the team allowed just 163 yards and 11 first downs while forcing one turnover, and while the competition in the 52-3 victory wasn't very high, it's set them off for a good 2025 season with a dominant start.

Is Duke ready to cause a monumental upset?

The Duke Blue Devils aren't known as a football school, but in recent years they've been highly competitive, with the team going 9-4 a season ago before a tight bowl loss to the No. 14-ranked Ole Miss. Unlike Illinois, they've turned to a new QB and a new offense in 2025, with sophomore transfer Darian Mensah expected to lead this team to plenty of success this season with his very balanced game.

That game was on full display in a week one win when the offense put up a ridiculous 548 yards, with Mensah throwing three touchdowns and no picks as they managed to rush for 159 yards as well. Defensively, the team limited FCS Elon to just 112 passing yards on the day as they started the season with a bang in their 45-17 victory.

Illinois vs. Duke pick

Spread Pick for Illinois vs. Duke

  • Illinois -3 (5 units)

While the line is just a tight -3, it's easy to see why Illinois comes into this one as a favorite, as the team was absolutely dominant in week one, and with the majority of the offense returning from a successful 2024 campaign, there are few reasons to doubt the No. 11-ranked team in the country. While Duke looked good last week, it came against an FCS team in Elon.

This will be the first matchup between Illinois and Duke, and while the home team should put up a good fight, their team just isn't quite as balanced as the Fighting Illini, who are ready to make a statement and prove they belong in the top-10 this season.

Over/Under Pick for Illinois vs. Duke

  • Over 49.5 (4 units)

A week ago, both of these teams dominated offensively, with over 440 yards on the day for each offense, and while the defense will stifle some of that, this matchup should see plenty of points put up on the board.

For the Fighting Illini, the over sits at 5-5 in their last 10 overall games, with 38 or more points scored in four of their past five, and for the Blue Devils, the over also comes in at 5-5 in their past 10, with six of their last seven overall seeing a total of 50 or more points scored.

Keep it simple, take the over.
